Like the open E chord, Esus2 uses the low E, B, and high e strings open. The fretted notes add the F# (the 2nd) that gives the chord its suspended colour.

Play E major, then Esus2, then back. The F# in Esus2 wants to rise to the G# of E major — that pull is the heart of a suspended chord.
